چکیده

Therapeutic indications and drug side-effects are both measureable human behavioral or physiological changes in response to the treatment. In modern drug development, both inferring potential therapeutic indications and identifying clinically important drug side-effects are challenging tasks. Previous studies have utilized either chemical structures or protein targets to predict indications and side-effects. In this study, we compared indication prediction using side-effect information and side-effect prediction using indication information against models using only chemical structures and protein targets. Experimental results based on 10-fold cross-validation, show that drug side-effects and therapeutic indications are the most predictive features for each other. In addition, we extracted 6,706 statistically highly correlated disease-side-effect pairs from all known drug-disease and drug-side-effect relationships. Many relationship pairs provide explicit repositioning hypotheses (e.g., drugs causing postural hypotension are potential candidates for hypertension) and clear adverse-reaction watch lists (e.g., drugs for heart failure possibly cause impotence). All data sets and highly correlated disease-side-effect relationships are available at http://astro.temple.edu/~tua87106/druganalysis.html.

Dydruma: A Dynamic Drug Map for Exploring the Associations Among Drug Therapeutic Indications and Side-Effects

Inferring potential therapeutic indications and identifying clinically important side-effects are both important and challenging tasks in modern drug development. Previous studies have utilized drug chemical structures and protein targets to construct predictive models for both tasks. According to our study, the drug therapeutic information itself is highly predictive for side-effects, and drug...

Characterizing the Network of Drugs and Their Affected Metabolic Subpathways

A fundamental issue in biology and medicine is illustration of the overall drug impact which is always the consequence of changes in local regions of metabolic pathways (subpathways). To gain insights into the global relationship between drugs and their affected metabolic subpathways, we constructed a drug-metabolic subpathway network (DRSN). This network included 3925 significant drug-metaboli...

Computational drug repositioning based on side-effects mined from social media

Drug repositioning methods attempt to identify novel therapeutic indications for marketed drugs. Strategies include the use of side-effects to assign new disease indications, based on the premise that both therapeutic effects and side-effects are measurable physiological changes resulting from drug intervention. Drugs with similar side-effects might share a common mechanism of action linking si...

The relation between Thiopurine Methyl Transferase activity and efficacy and side effects of Azathioprine in patients with Pemphigus Vulgaris

Background and aim: Azathioprine is the most widely used immunosuppressive agent as an adjunct to corticosteroids in the treatment of pemphigus vulgaris (PV). Thiopurine methyl transferase (TPMT) is a key enzyme in azathioprine metabolism and a genetic polymorphism controls its activity in human tissue. TPMT activity can provide a rational basis to determine suitable dose of azathioprine, theor...
